Test Principle:γ-Aminobutyrate transaminase (GABA-T , EC 2.6.1.96) is a pivotal enzyme in the γ-aminobutyric acid (GABA) pathway , catalyzing GABA’s degradation. GABA plays a fundamental role in the central nervous system by maintaining emotional stability , promoting sleep , regulating movement , and preventing epilepsy , primarily through its inhibitory function. Consequently , GABA-T serves as a critical regulator of neuronal excitability and a key determinant of GABA concentration. In brain tissue , the GABA-T catalyzed reaction utilizes α-ketoglutarate as an amino-group acceptor , converting GABA into succinic semialdehyde and glutamate. Subsequently , succinic semialdehyde dehydrogenase oxidizes succinic semialdehyde to succinate , reducing NAD? to NADH in the process. This reduction forms the basis for the activity assay: NADH has a characteristic absorption peak at 340 nm , and the rate of its increase is directly proportional to GABA-T activity.
