Testprinzip: Lyophilized Amebocyte Lysate (LAL) is made from amoebocytes of Tachypleus tridentatus. It contains factor C, factor B, coagulase, and coagulinogen. Under suitable conditions, endotoxin activates C factor, followed by a series of enzymatic reactions, activating coagulinogen to form coagulase which cleaves the chromogen supplied in this kit to produce a yellow p-nitroaniline (pNA, λmax = 405nm). The amount of pNA produced is positively correlated with the endotoxin concentration over a certain period of time. Meanwhile, pNA can react with the azo reagent to produce a rose-red end product (λmax = 545nm), by which the endotoxin concentration of the sample can be determined and the interference of the sample color with the absorbance at 405 nm can be avoided as well.
