Test Principle:
The hydroxyl radical is a highly toxic and harmful free radical produced by the human body during metabolism. It can oxidize substances such as carbohydrates , amino acids , proteins , and nucleic acids in tissues , leading to oxidative damage and destruction , and resulting in cell necrosis or mutation. The hydroxyl radical scavenging capacity is one of the key indicators of a sample’s antioxidant activity and is widely applied in studies on plant stress tolerance , antioxidant health products , and pharmaceuticals.



In the Fenton reaction , Fe²⁺ and H₂O₂ generate hydroxyl radicals while oxidizing Fe²⁺ to Fe³⁺. Salicylic acid chelates with Fe³⁺ to form [Fe(C₆H₄O₂)(H₂O)₄]⁺ (purple complex) , which exhibits an absorption peak at 520 nm. By measuring the absorbance at 520 nm , the hydroxyl radical scavenging ability of the sample can be evaluated.
