Test Principle:
Levodopa (L-DOPA, EC 200-445-2) is a naturally occurring aromatic amino acid and also the direct precursor of dopamine. It is widely found in medicinal plants such as broad beans and velvet beans. As a classic and effective drug for treating Parkinson's disease, L-DOPA can smoothly cross the blood-brain barrier and be converted into dopamine in the brain to replenish the deficiency of neurotransmitters. Its medicinal value and natural properties have attracted significant attention. Determining its content is crucial for drug quality control, plant screening, and biochemical research.

Under acidic conditions, L-DOPA is oxidized by nitrous acid to form dopachrome quinone. After alkalinization, dopachrome quinone cyclizes to form colorless leucodopachrome. Catalyzed by sodium molybdate, it is oxidized to orange-red dopachrome, which exhibits characteristic absorption at 500 nm. The absorbance is directly proportional to the content of L-DOPA. By measuring the absorbance value at a wavelength of 500 nm, the content of L-DOPA in the sample can be calculated.
