Testprinzip: Neutral Red (NR) is a pH indicator, being red under acidic conditions and changing from red to yellow as pH rises from 6.8 to 8.0. In live cells, NR accumulates in lysosomes with acidic environment, and therefore stains live cells in red. After cell fixation, the acidic environment in lysosome is destroyed and the lysosome can not be stained by NR. Instead, cell nuclei are stained in dark red by NR that binds to acidic DNA and RNA in large amounts in nucleus, while cytoplasm is stained in light red.
