Test Principle:
Phospholipases D (PLD) are crucial regulatory enzymes in the metabolic process of organisms, closely related to membrane lipid metabolism, cell signal transduction, cell proliferation and differentiation, as well as stress responses. The activity of phospholipase D is an important indicator for evaluating the functional state of the cell membrane and the cell signaling pathways.

Phospholipase D catalyzes the hydrolysis of phosphatidylcholine to produce phosphatidic acid and choline. Choline is catalyzed by choline oxidase to produce betaine and hydrogen peroxide. Hydrogen peroxide reacts with a fluorescent probe under the action of peroxidase to form a strong fluorescent product. The fluorescence intensity of this product at an excitation wavelength of 535 nm and an emission wavelength of 587 nm is proportional to the content of H2O2.

Reagents and Equipment Required but Not Provided:Fluorescent microplate reader, balance, benchtop centrifuge, water bath/thermosetting incubator/metal bath, adjustable pipette, 96-well black microplate (fully black, 1 piece included in the box), sealing film (2 sheets included in the box), mortar/lymphomizer/cell ultrasonic disruption instrument, ice and distilled water.
