Test Principle:Tyrosinase ( Tyrosinase ; EC 1.14.18.1 ) is a key rate-limiting enzyme in the process of melanin synthesis. Its inhibitors have important research value in cosmetic whitening, anti-browning of fruits and vegetables, and treatment of certain diseases. Based on the principle of enzyme reaction kinetics, this kit calculates the inhibition rate by detecting the absorbance change of the product at a specific wavelength, which is suitable for screening and evaluating the inhibition rate of tyrosinase inhibitors. L-tyrosine generates dopa under the action of tyrosinase, and dopa is catalyzed by tyrosinase to generate dopaquinone. Dopamine has an absorption peak at 475 nm. After adding tyrosinase inhibitor, the formation of dopaquinone can be inhibited, and the tyrosinase inhibition rate can be calculated accordingly.
