Test Principle:
Carnitine palmitoyltransferase (CPT) is divided into CPT-I and CPT-II. CPT-I is a rate-limiting enzyme in the β-oxidation of fatty acids. It is located on the outer membrane of mitochondria and catalyzes the transfer of long-chain fatty acids from acyl-CoA to carnitine and then from the cytoplasm into the mitochondria, and further β-oxidation is catalyzed by CPT-II located on the inner membrane of mitochondria. The dysfunction of carnitine palmitoyl transferase may lead to fatty acid metabolism disorders, which in turn cause a series of metabolic diseases.

Under the action of carnitine acyltransferase (CPT-I), carnitine and palmitoyl coenzyme A produce acylcarnitine, and release free coenzyme A (COA-SH). After reacting with Ellman reagent DTNB, yellow TNB is produced. The activity of CPT-I is calculated by the change of its absorption peak (412 nm).
