Fatty acid β-oxidation is a biochemical process primarily occurring in the mitochondrial matrix of cells. It involves the stepwise breakdown of long-chain fatty acids to generate acetyl-CoA, ultimately yielding energy. The fundamental steps of fatty acid β-oxidation comprise four major phases: activation of acyl-CoA, acyl-CoA entry into mitochondria, the β-oxidation cycle of acyl-CoA, and complete oxidation of acyl-CoA. This process converts fatty acids into substrates for the tricarboxylic acid (TCA) cycle while generating ATP within mitochondria, thereby supplying cellular energy.
In the presence of substrates and cofactors, NADH is generated during fatty acid β-oxidation. This NADH reacts with an electron-coupling agent and a chromogenic reagent to form an orange-red compound exhibiting a characteristic absorption peak at 485 nm. This peak allows for the quantification of the fatty acid β-oxidation rate.
