Glutamate dehydrogenase ( EC 1.4.1.2 ) is the core hub connecting nitrogen metabolism and carbon metabolism, and its determination is of key significance in clinical diagnosis and basic research. In clinical practice, serum GDH activity is a highly specific indicator for assessing liver cell injury, especially for alcoholic liver disease and drug-induced liver injury. In the field of liver transplantation, GDH measurement can effectively determine the degree of ischemia-reperfusion injury and cell viability of transplanted liver. In addition, in the diagnosis of congenital metabolic diseases, GDH deficiency can lead to high blood ammonia and neurological abnormalities, and enzyme activity detection is the basis for diagnosis. In basic research, the mechanism of ammonia assimilation, amino acid metabolism and energy balance regulation can be further explored by measuring GDH activity, which provides a theoretical basis for the research of metabolic diseases and the development of new drugs.

L-glutamic acid and NADP + produce NH4 +, α-ketoglutarate and NADPH under the catalysis of glutamate dehydrogenase. NADPH has an absorption peak at 340 nm. By measuring the increase rate of absorbance at 340 nm, GDH activity can be measured.
