Test Principle:
Homocysteine (Hcy) is a non-proteinogenic amino acid and a sulfur-containing intermediate produced during methionine metabolism, primarily generated through the demethylation of methionine. Hcy maintains cellular methylation levels via the methionine cycle and serves as a rate-limiting substrate in the transsulfuration pathway for the synthesis of the antioxidant glutathione. Furthermore, hyperhomocysteinemia is an independent risk factor for cardiovascular diseases and stroke.

Oxidized Hcy in the sample is reduced to free Hcy using a reducing agent. Cystathionine-β-Synthase catalyzes the reaction between free Hcy and serine to form cystathionine. Cystathionine β-Iyase cleaves cystathionine to generate Hcy and pyruvate, thereby establishing a Hcy cycling amplification system. The generated pyruvate is subsequently converted to lactate by lactate dehydrogenase with the concomitant consumption of NADH to form NAD+. The Hcy content can be calculated according to the decrease rate of NADH at 340 nm
