Plant roots are active absorption and synthesis organs. Root growth and metabolism, namely root activity, directly affect plant shoot growth, nutritional status and yield, and is one of the important physiological indicators of plant growth. According to the theory of Sabinin et al., the absorption of solutes by plants has the characteristics of surface adsorption, and it is assumed that the adsorbed substances form a uniform monolayer layer on the surface of the root system. When the adsorption of solutes by the root system reaches saturation, the active part of the root system can further transfer the attached substances to the cells and continue to produce adsorption.

The detection principle of the kit takes methylene blue as adsorbent substance, and the adsorbed amount can be calculated according to the change of methylene blue concentration before and after adsorption. The concentration of methylene blue can be measured by colorimetry, and the absorbance at 660nm can be detected by spectrophotometer or microplate reader. It is known that the area covered by 1mg of methylene blue when forming a monolayer is 1.1m2. Based on this, the total absorption area of the root system can be calculated. From the amount of methylene blue adsorbed after saturation, the active absorption surface area of the root system can be calculated.
