Test Principle:Casein is a common substrate for a variety of proteases. When casein is labeled with FITC ( Fluorescein isothiocyanate ), the fluorescence of FITC is quenched due to fluorescence resonance energy transfer. When the protease catalyzes the hydrolysis of FITC-labeled casein substrate, the fluorescence quenching effect is destroyed and the fluorescence signal is enhanced. The fluorescence intensity at the excitation wavelength of 485 nm and the emission wavelength of 525 nm is proportional to the activity of the protease.
