'pyruvate phosphate dikinase (PPDK, EC 2.7.9.1) is a rate-limiting enzyme of the C4 pathway and the sedum acid metabolism pathway, which catalyzes the three-step reaction of ATP, pyruvate and Pi to produce phosphoenolpyruvate. This enzyme is mainly found in the chloroplast matrix of C4 plants and has an important regulatory role in photosynthetic function.
Measurement Principle:
The reverse reaction of PPDK catalyzes the generation of pyruvate by phosphoenolpyruvate (PEP), AMP and PPi to pyruvate, ATP and Pi, and lactate dehydrogenase (LDH) further catalyzes the generation of lactate and NAD+ by pyruvate and NADH, and the rate of NADH reduction is measured at 340 nm, from which PPDK activity can be calculated.
