Test Principle:Ribonuclease III (RNase III) is a double-stranded RNA (dsRNA)-specific ribonuclease derived from Escherichia coli. It is incapable of hydrolyzing DNA or single-stranded RNA. In the presence of manganese ions (Mn2?),RNase III specifically cleaves long dsRNA into small interfering RNAs (siRNAs) of approximately 18-25 base pairs (bp). These siRNAs possess a 3’-hydroxyl terminus with 2 to 3 overhanging bases. The cleavage products are structurally similar to those generated by Dicer,and this enzyme is widely applied in researches including RNA interference,gene expression regulation,gene silencing and target validation.
