Introduction: As a type of food additive, sulfites exert multiple functions including bleaching, preservation, antioxidation, bacterial growth inhibition and enzymatic reaction regulation, and are widely applied in the food industry. However, excessive sulfur dioxide may cause certain damage to the human respiratory and digestive systems, with particularly severe harm to asthma patients. Meanwhile, it can induce somatic mutation and even possess carcinogenic potential. The detection of sulfites (or sulfur dioxide, SO?) has been extensively applied in fields such as food safety, disease research and environmental monitoring.

Principle: Sulfites react with formaldehyde to generate hydroxymethylsulfonate (HMS). In a strongly acidic medium, this intermediate undergoes a specific color reaction with pararosaniline hydrochloride (PRA), forming a purplish-red complex with a maximum absorption wavelength of 585 nm. Quantitative analysis of sulfites (or SO?) can be realized by determining the absorbance at this wavelength.

Self-prepared Instruments and Supplies: Spectrophotometer, 1 mL glass cuvette, analytical balance, low-temperature centrifuge, water bath/constant temperature incubator, adjustable pipette, mortar/homogenizer, ice and distilled water.
