Test Principle:Oxidized glutathione (GSSG) is the oxidized form of glutathione (GSH), also known as dithioglutathione, which is oxidized by two molecules of glutathione. GSSG is reduced to GSH by glutathione reductase, so most of the body exists in a reduced form. Determination of intracellular GSH and GSSG content and GSH/GSSG ratio can well reflect the redox state of cells. The endogenous GSSG in the sample was completely reduced to GSH by glutathione reductase (GR), and the total glutathione content in the system was the sum of GSH and GSSG. GSH can react rapidly with the chromogenic reagent 5,5 ’ -dithiobis (2-itrobenzoic acid, DTNB) to form yellow products 5-thio-2-nitrobenzoic acid (TNB) and GSSG.TNB has a characteristic absorption peak at 412 nm, so the total glutathione content in the sample can be calculated. The sample was treated with 2-vinylpyridine in advance to remove endogenous GSH, and the color reaction was performed according to the above steps to determine the content of GSSG.Finally, the GSH content was indirectly obtained by the difference between the total glutathione content and the GSSG content.
