Xanthine oxidase (XOD,EC 1.17.3.2) is one of the key enzymes of nucleotide metabolism and is mainly distributed in mammalian heart,lung,liver and other tissues. When liver function is impaired,XOD is released into serum in large amounts,which has specific significance for the diagnosis of liver damage.

XOD catalyzes the oxidation of hypoxanthine to xanthine and subsequently catalyzes the oxidation of xanthine to uric acid,while producing hydrogen peroxide.Hydrogen peroxide reacts with a fluorescent probe to generate a strong fluorescent product. The fluorescence intensity at an excitation wavelength of 535 nm and an emission wavelength of 587 nm is proportional to the concentration of hydrogen peroxide. The XOD activity can be calculated by measuring the generated hydrogen peroxide.
